1. Sample treatment

(1) Mix 60 µL of the water sample with 60 µL of the reconstitution solution thoroughly, and keep for use.

(1) Remove the skin and select parts with minimal fat content. Use a grinder to homogenize the sample.

(2) Place 4g±0.1g of defatted sample in a 15mL centrifuge tube.

(3) Add 2 mL of Reagent 1 to the above centrifuge tube and shake vigorously for 2 min. Then sequentially add 150 µL of Extractant 1, 4 mL of acetonitrile, and 2 mL of n-hexane. Tighten the cap and shake vigorously for 2 min, followed by centrifugation at 4000 r/min for 5 min at room temperature.

(4) At this point, the bottom of the centrifuge tube contains the homogenized tissue sample. From bottom to top, three liquid layers are formed. Carefully transfer 2 mL of the clear middle layer into a 5 mL centrifuge tube, add 100 µL of Oxidant, gently shake in parallel to mix for 30 seconds, and dry at 65 °C using a nitrogen evaporator or a blow dryer.

(5) Add 0.3 mL of MG reconstitution solution to the dried centrifuge tube. Pipette up and down repeatedly for 1 minute to fully reconstitute the solid residues at the bottom and on the inner wall of the tube. Withdraw 120 µL (approximately 5-6 drops) of the solution; What we get is test fluid.
